Zebrafish, in particular, has become an attractive model system to study circadian rhythms because of its advantages for genetic analyses, the optical clarity of its embryos and low maintenance cost.
OrpheX screening assay to identify selective NCR or DCR compounds.
LT embryos are arrayed in multi-well plates. The plates are kept at NCR (DL) or DCR (DD) conditions in a temperature-controlled water system at constant temperature.
The assay allows selecting compounds that modulate clock protein activity specifically under DCR.
The drawing illustrates that LT larvae have impaired protein activity under DCR when treated with vehicle, drug 1 activates only larvae raised under NCR condition, drug 2 activates larvae in both conditions whereas drug 3 only activates larvae raised under DCR condition.
OrpheX applied the assays successfully and identified DCR selective compounds.
